Digital Single DNA Molecule Detection By CRISPR/Cas12a: Towards Targeting Single CpG Site Methylation

Description

The focus of this talk will be on CRISPR-Cas biosensing. Previously, it was shown that hypermethylation of specific genes could be related to several cancer types. With the upcoming field of liquid biopsies and the conformation that these hypermethylated DNA sequences could be found in urine, the development of novel biosensors for fast and reliable detection of these sequences becomes of interest. We developed a CRISPR/Cas in vitro diagnostic platform that can be used to detect single methylation points in the DNA with a simple fluorescence read-out. To move towards clinical applications, we present our work on single molecule CRISPR methods that enable the detection of target DNA molecules within the femtomolar range, potentially enabling amplification-free detection of hypermethylated genes in patient samples.
